Cross protection by heat and cold shock to lethal temperatures in Clostridium perfringens BJM

A heat shock pre-treatment of Clostridium perfringens provides protection against lethality from subsequent exposure to cold shock and vice versa. Heat shocked cells were more cold tolerant (one log reduction in 65 min) than control cells (one log reduction in 34 min). On the other hand, cold-shocked cells were more thermotolerant (D55= 17 min) than the control (D55= 6.5 min). The addition of chloramphenicol (an inhibitor of protein synthesis) in the experiments indicates that synthesis of new proteins was necessary for cross protection between both treatments.

Effect of high temperature on germination of four legumes from a forest-grassland mosaic in Southern Brazil Biota Neotropica

Passage of a fire can break dormancy of the seeds of many plant species in fire-prone ecosystems. This response to fire is especially well known for the Fabaceae family, but has been poorly studied in Southern Brazil. We collected seeds of four Fabaceae species present in grasslands-forest mosaics of Southern Brazil: Mimosa bimucronata, Desmodium barbatum, Sesbania virgata and Collaea stenophylla. Seeds were exposed to different heat treatments (exposure to 60° and 80 °C for 5 minutes, to 100 °C for 2 minutes, control without heat treatment); not all species were tested in all treatments. After the treatment, the seeds were kept in a germination chamber with a light period of 12/12 hours and temperature of 25 °C. Germinated seeds were counted every 2 days....

Heat-shock response and temperature resistance in the deep-sea vent shrimp Rimicaris exoculata ArchiMer

The shrimp Rimicaris exoculata swarms around hydrothermal black smoker chimneys at most vent sites along the Mid-Atlantic Ridge. This species maintains close proximity to the hydrothermal fluid, where temperatures can reach 350degreesC and steep thermal and chemical gradients are expected. We performed in vivo experiments in pressurized aquaria to determine the upper thermal limit [critical thermal maximum (CTmax)] of R. exoculata and to investigate some characteristics of the shrimp stress response to heat exposure. These experiments showed that the shrimp does not tolerate sustained exposure to temperatures in the 33-37degreesC range (CTmax). Lysozyme gene expression and hemocyte behaviour in the Mediterranean mussel, Mytilus galloprovincialis, after injection of various bacteria or temperature stresses ArchiMer

The aim of the present study was to evaluate the expression of the Mytilus galloprovincialis lysozyme gene in different in vivo stress situations, including injection of bacteria Vibrio splendidus LGP32, Vibrio anguillarum or Micrococcus lysodeikticus, as well as heat shock at 30 C and cold stress at 5 C. Injection of V. splendidus LGP32 resulted in: (i) a general down-regulation of lysozyme gene expression, as quantified by Q-PCR; (ii) reduction in the number of circulating hemocytes; (iii) decrease in the percentage of circulating hemocytes expressing lysozyme mRNA which was now restricted to only small cells, as observed by ISH; and (iv) accumulation of hemocytes expressing lysozyme in the muscle sinus where injection took place. Este documento descreve o modelo molecular da proteína XF2234 e a análise preliminar da sua estrutura. O modelo foi construído por modelagem por homologia (ou modelagem comparativa) com a estrutura cristalográfica de uma proteína small heat shock de Triticum aestivum (trigo). A análise da estrutura tridimensional da proteína XF2234 tem o objetivo de contribuir para aumentar o conhecimento sobre o papel biológico das smHSPs, necessário para o combate à CVC.

Mortality among juvenile Crassostrea gigas has been occuring during the summer months since 1995 and is now considered to be a real problem for French oyster culture. The aim of this study was to standardise the stress that could be applied to Crassostrea gigas spat. In initial experiments in 1998, the weight of individual spat and the duration of a heat shock at 50°C in freshwater, adapted for oysters of between 500 and 2000 mg, were built into a model to obtain 50% mortality. However, the precision was low (50%) and will need to be improved in the future. Prolonged exposure of human embryonic stem cells to heat shock induces necrotic cell death Biocell

We investigated the effects of prolonged heat shock treatment on human embryonic stem cell (hESC) viability. The hESC viability steadily declined with longer exposure to heat shock treatment (43ºC). After 4 h of exposure to heat shock at 43ºC, only 56.2 ± 1.5% of cells were viable. Viability subsequently declined to 37.0 ± 3.3% and 3.5 ± 0.7% after 8 h and 16 h, respectively of heat shock treatment at 43ºC. Transmission electron micrographs showed that the morphology of the dead/dying cells after heat shock treatment was characteristic of cellular necrosis with an uncondensed chromatin and a non-intact plasma membrane.
